초록
Objective : Women have a big physical change since they have menopause. and during this period, social and mental enviroments cause a lot of stress to women, so the incidence rate of depression is higher than any other time. The depression is similar to the Kiui-jeung(氣鬱證), so Soyo-San is used clinically as a model in Oriental medicine. Thus this experiment is made for test whether Soyo-San have an antidepressant effect in menopausal depression or not. Method : The female Sprague-Dawley(200±10g) rats were used, the temperature was controlled within 20±2℃ and water and food were not limited supposing the day and night 12hours each. After Pentobarbital sodium(50㎎/㎏) was injected into intrapentoneal to anesthetize it, we made 1㎝ cut on the abdominal of the rat to remove the ovaries and started to dose Soyo-San extract divided low dose group(Soyo-San extract 280㎎/㎏/day medicating group, as follow indicating Sample A group) and high dose group(Soyo-San extract 560㎎/㎏/day medicating group, as follow indicating Sample B group) orally in 3weeks after, we are not measured the locomotor activity for rule out possibility decreased the immobility duration and frequency because nonspecific activity increase, until we experiment the forced swim test as a behavior test that we could prospect an antidepressant effect Result : 1. The immobility duration in the forced swim test decreased dose dependently as we medicated Soyo-San. 2. The immobility frequency in the forced swim test decreased dose dependently as we medicated Soyo-San. 3. One of the active behavior, the climbing frequency increased significantly in both Sample A group(P<0.02) and Sample B group(P<0.01) compared to the control group dose dependently. 4. One of the active behavior, the head twitch frequency increased significantly in Sample B group(P<0.02) compared to the control group dose dependently. 5. One of the active behavior, the swimming frequency is a little inclined to increase dose dependently as we medicated Soyo-San. 6. The sum of active behavior frequency that binds the climbing, head twitch and swimming's frequency of all is inclined to increase dose dependently as we medicated Soyo-San. 7. The sum of a initial hour locomotor activity decreased significantly in Sample A group(P<0.05) compared to the control group. 8. The basal locomotor activity adding up the sum of a next hour was a little inclined to decrease in Sample B group but there was not much difference. Conclusion : as a result of this experiment, the possibility of Soyo-San is prospected as a medical treatment to cure women's menopausal depression inferred to the facts that Soyo-San decrease the immobility and increases the active behavior such as the climbing of rats.
