Heat shock protein 70 mRNA expression by Intraperitoneal kainic acid injection

초록

The effects of systemic administration of kainic acid on heat shock protein 70 mRNA expression in the rat brain was studied by in situ hybridization histochemistry and RNA blot analysis. The result of in situ hybridization histochemistry showed that the level of hsp 70 mRNA began to increase 3hr after the administration of kainic acid in a convulsant dose( 10mg/kg i.p.), peaked at 12hr after injection and returned to basal level at 96hr after injection. The result of RNA blot analysis was well matched with that of in situ hybridization histochemistry. The result of RNA blot showed only the temporal pattern of the change of hsp 70 mRNA, but the result of in situ hybridization histochemistry showed not only the temporal but also the spatial pattern of the changes. The spatial pattern of the kainic acid induced expression of hsp 70 mRNA in the rat brain was that kainic acid receptor rich areas(CA 3/4 areas of hippocampus, amygdala, piriform cortex and thalamus) showed earlier, higher and more steady expression than other areas. Then, temporal and spatial pattern of hsp 70 mRNA expression suggest that the expression of hsp 70 by the kainic acid may be due to direct effect on the kainic acid receptor not by the indirect effect of the kainic acid induced convulsion.
