Effects of genistein, genistin, and daidzein on preterm Drosophila model and human cervical epithelial cells

Objective Sex hormones play an important role in reproduction. Our previous study confirmed that the ratio of ER and PR was increased in the cervix of mouse preterm birth model. Genistein, genistin, and daidzein have been reported to regulate ER and PR expression. In this study, the effect of genistein, genistin, and daidzein on Drosophila sex hormone level, egg production and gestational period as well as the effect on ER and PR expression in the cervix epithelial cell line ME-180, were investigated. Study Design ETH expression was analyzed using quantitative PCR in each group of Drosophila melanogaster. In order to confirm the effect of genistein, genistin, and daidzein on Drosophila egg production, each standard (lead nitrate, genistein, genistin, and daidzein) solution was added. The treatment of lead nitrate for infertile modeling was used to suppress egg production. Egg production was confirmed by counting. The gestational period from mating and egg production was measured every fixed time. ME-180 cells were seeded in a 60 mm dish at 5ⅹ10 5 , treated with daidzein at respective concentrations, and RNA was extracted after 24 hr. The expression of ER and PR was confirmed by real-time PCR. Results The expression of Drosophila ETH was increased by genistein (p = 0.483), genistin (p < 0.001), and daidzein (p = 0.216). Egg production and gestational period were decreased in Drosophila fed a medium containing lead, however they were increased in Drosophila fed media treated with genistein (p = 0.020), genistin (p < 0.001), and daidzein (p = 0.269). Next, we checked ER and PR expression and ER/PR ratio in ME180 cells treated with daidzein. As a result, it was confirmed that ER level (p = 0.014) decreased, PR level (p < 0.001) increased, and ER/PR ratio (p < 0.001) increased at a specific concentration. Conclusion Genistein, genistin, and daidzein increase fertility and gestational period in Drosophila. The treatments were effective in reducing the ER/PR ratio and would be potential therapeutic targets for preventing preterm birth.
