MAX Eraser: An affordable and simple reagent for antibody removal cycle in multiplex immunohistochemistry

Immunochemical techniques are widely used to identify and localize antigens in fixed specimens. Recent advances in multiplex imaging techniques have enabled the detection of multiple proteins from a single specimen, but these techniques often require expensive devices and complex experimental steps. Here, we show the development of MAX Eraser, a chaotropic reagent-based solution that disrupts antibody-antigen interactions at room temperature without significant specimen deformation. The optimized conditions allow up to 10 cycles of immunohistochemistry → imaging → antibody removal, making it suitable for multiplex imaging. MAX Eraser can be used in conventional and super-resolution imaging applications with a wide range of specimens such as cultured cells, formalin-fixed and paraffin-embedded or frozen tissue sections, and thick 3D specimens. We propose that MAX Eraser is a simple and affordable reagent for multiplex imaging. © 2025 Elsevier B.V., All rights reserved.
