Immunohistochemical Study of the Distribution of Inositol Phospholipid-Specific Phospholipase C-β1 in the Rat Brain

초록

Inositol phospholipid-specific phospholipase C(PLC) plays a central role in the signal transduction pathway because it catalyzes the hydrolysis of phosphatidylinositol 4,5-bisphosphate generating two second messenger molecules, inositol 1,4,5-trisphosphate and diacylglycerol. The distribution of the inositol phospholipid-specific phospholipase C-β1(PLC-β1) was observed in the rat brain by immunohistochemical staining with monoclonal antibody directed against PLC-β1. PLC-β1 showed ubiquitous pattern of distribution in the rat brain. Generally forebrain showed higher immunoreactivity than hindbrain. PLC-β1 immunoreactive staining was densely observed in the striatum, cerebral cortex, dorsal lateral septal nucleus, medial subdivision of the mediodorsal thalamic nucleus, anteromedial thalamic nucleus, basolateral amygdala nucleus, CA1 and other areas of hippocampus, subiculum, dentate gyrus, facial nucleus, and trigeminal nucleus. PLC-β1 immunoreactivity was mainly localized in the neurons, and the glial cells were not immunostained. Neuronal perikarya and the dendritic processes had the most clear immunoreactive labeling. In addition, the neuronal nuclei also showed a clear immunoreactivity in the thalamic, intracerebellar, and brainstem nuclei. The widespread distribution of PLC-β1 in the neurons of the rat brain suggests that this enzyme may play an important part in the common system of signal transduction used universally by the neurons of the brain.
