Clinical implication of tissue-resident memory T cells in psoriasis

Tissue-resident memory T cells (TRM) represent a subset of non-circulating memory T cells that play a vital role in immune surveillance and the protective response against reinfection of exogenous pathogens. However, a pathogenic population of TRM has been implicated in the pathogenesis of chronic skin inflammatory diseases such as psoriasis. The natural course of psoriasis, characterized by long-term persistence and repetitive recurrence of lesions in the same region, highlights the importance of TRM in psoriasis. Even before the actual discovery of TRM, there has been evidence that lesional T cells, rather than skin-homing T cells, are important in psoriasis pathogenesis. Subsequent key experiments confirming the existence and characteristics of TRM in the skin greatly improved our understanding of their role in psoriasis. In terms of treatment, several treatment methods have shown potential for decreasing TRM counts, and some can even reduce the frequency of TRM in psoriasis lesions. However, TRM are known to remain numerically stable for a considerable time. Therefore, early intervention to decrease the reservoir of TRM is suggested to allow disease modification in psoriasis. In this review, we will review the role of TRM and discuss the treatment implications of TRM in psoriasis.
