인체 지방세포에서 유래된 줄기세포와 폴리락타이드/글리콜라이드 공중합체(PLGA)복합체를 이용한 백서의 두개골 결손의 골형성 유도

초록

The present study was aimed to assess the effect in bone regenerating capability of human adipose-derived stem (ASC) cells cultured in osteogenic media with the Poly lactide-co-glycolic acid (PLGA) and implanted in a nude rat calvarial defect. Twenty seven nude rats were randomized into three different groups (n=9) and these groups were divided into 9 or 3subgroups according to pre-cultured periods (day 1 (group A), day 7 (group B), and day 14 (group C) days) before implantation. By radiodensitometric analysis, ASC cell-loaded implants (group 2 and 3) except 1 day pre-culture groups demonstrated much better results than cell unloaded implants(group 1) and the implant precultured in osteogenic media for 14 days demonstrated to generate the best results.For histomorphometric analysis, Group 2 and 3 all demonstrated maximal bone formation at the 2 weeks precultured time point, filling 35-72% of the area of the defect. Noticeably, group 3 had robust bone tissue regeneration than group 2. These data show that Group 2 and 3 heal critical-size skeletal defects. Importantly, differentiated osteogenic ASCs showed a better critical defect repair. Human adipose derived stromal cells can therefore be used as the seed cells to construct tissue-engineered bone.
