Abstract
성숙한 숫 고양이 뇌에서 Inositol 1,4,5-trisphosphate 3-kinase (IP3K)의 분포를 단일 세포군항체를 이용한 면역조직화학적 방법으로 조사하였다. 그 결과 소뇌의 조롱박신경원, 해마의 CA4다형세포, 외측중격핵 및 꼬리핵과 조가비핵의 중간크기의 신경세포에서 가장 강한 면역반응을 관찰할 수 있었고, 대뇌피질의 피라밋세포와 CA1, CA3 피라밋세포 및 해마 능선부분의 피라밋세포에서 중등도의 면역반응을 관찰할 수 있었으며, 또 신경연접요소인 가지돌기가시로 보이는 점상의 면역반응도 관찰할 수 있었다. 치아이랑의 과립세포, 편도체 및 담장의 신경세포에서는 약한 면역반응을 관찰할 수 있었다. 한편 소뇌의 과립세포와 소뇌핵, 시상과 시상하부, 뇌간의 신경세포 및 신경교세포와 혈관내피세포에서는 면역반응을 인정할 수 없었다. 결론적으로 대뇌피질, 해마체, 외측중격핵 및 소뇌의 조롱박신경원 등 신경연접의 유연성이 높은 부위로 알려진 중추신경계의 특별한 부위에 IP3K가 풍부하게 존재하는 특이한 분포양상을 관찰할 수 있었다.
Inositol 1,4,5-trisphosphate 3-kinase (IP3K) phosphorylates Inositol 1,4,5-trisphosphate (InsP3) to produce inositol 1,3,4,5-tetrakisphosphate (InsP4) through agonist stimulation. InsP4, is emerging as a second messenger which controls Ca2+ homeostasis or regulates InsP4-mediated K+ channel. Although the role of InsP4 is not yet clearly understood and its receptor protein has not been clearly identified, InsP3 and InsP4 and their receptors may have a significant role in Ca2+-induced intracellular signal transduction mechanism. This study was attempted to investigate the IP3K distribution in the cat brain by immunohistochemical study using monoclonal antibodies against IP3K. The IP3K immunoreactivity was found in the neurons of the cerebral cortex, hippocampus, and cerebellum which were well known as the area of high synaptic plasticity. The highest immunoreactivity was observed in the perikarya and dendritic tree of Purkinje cells in the cerebellum, polymorphic cells in the CA4 area of hippocampus, neurons in lateral dorsal septal nucleus, and caudate-putamen. Moderate high immunoreactivity was found in the perikarya and dendritic field of pyramidal neurons in the cerebral cortex, pyramidal neurons of the CA1 to CA3 areas of hippocampus, subiculum, and low immunoreactivity was found in the granule cells of dentate gyrus, amygdaloid nucleus, indusium grisium. The reactivity of cerebellar granule cells, deep cerebellar nuclei, thalamus, hypothalamus, brain stem, and white matter tracts was almost negative. There was no obvious immunoreactivity in glial cells and endothelial cells. This finding may suggest a specialized role for IP3K in the brain.
